How It Works

The HIC columns are packed with phenyl-modified agarose, which selectively retains hydrophobic impurities:​

• Unbound Fluorescent Dyes: Aromatic dyes like FAM and HEX bind to phenyl groups via π-π stacking interactions, effectively removing them from the sample.​

• Salts and Small Molecules: These enter the agarose pores and are retained, while larger oligonucleotides pass through unimpeded.​

This method ensures that labeled oligonucleotides are not retained by the column and are recovered efficiently.​
